KADAR MDA SPERMATOZOA SETELAH PROSES PEMBEKUAN

Abstract


Tujuan penelitian ini adalah untuk mengetahui efek lama pembekuan semen terhadap kadar MDA spermatozoa dan persentase viabilitas spermatozoa. Sampel yang digunakan berupa  semen beku sapi Limousin dengan lama pembekuan 2 bulan dan 3 bulan yang dikoleksi dari BBIB Singosari. Semen beku yang telah dithawing diencerkan dengan Phosphate Buffer Saline (PBS) kemudian disentrifugasi untuk memisahkan spermatozoa dan seminal plasma. Kadar MDA spermatozoa diukur dengan menggunakan uji TBA dan spektrofotometer. Pengamatan viabilitas spermatozoa dilakukan dengan menggunakan pewarnaan eosin-negrosin. Data berupa kadar MDA spermatozoa dan persentase viabilitas dianalisis dengan uji-t berpasangan dan korelasi pearson menggunakan spss 16.0. Hasil analisis menunjukkan bahwa kadar MDA spermatozoa pada lama pembekuan 2 bulan (0,0777±0,008 nM) dan 3 bulan (0,0919±0,016 nM) tidak berbeda nyata (p<0,05). Terdapat korelasi negatif antara persentase viabilitas dan kadar MDA pada spermatozoa.
