AMPLIFIKASI GEN COI (Cytochrome C Oxidase Subunit I) PADA HOLOTHUROIDEA YANG DIKOLEKSI DARI KEPULAUAN RAJA AMPAT
Abstract
ABSTRAK
Kepulauan Raja Ampat mempunyai keanekaragaman hewan laut yang tinggi, salah satunya Timun Laut (Holothuroidea). Tujuan dilaksanakan penelitian ini, yakni mengamplifikasi gen COI pada Holothuroidea yang dikoleksi dari Kepulauan Raja Ampat. Metode yang digunakan pada penelitian ini yakni total DNA diisolasi dari jaringan menggunakan gSYNCTM DNA Extraction Kit GS100 (Gene Aid), kemudian DNA spesimen UNP 106 diamplifikasi berdasarkan gen COI menggunakan susunan sekuen forward COI yakni 5’-GGCATCCAGAAGTTTACATCCT-3’ dan sekuen reverse yakni 5’-TTGGAGATAGGCTTCTGTGGA-3’. Hasil penelitian menunjukkan bahwa spesimen UNP 106 berhasil diamplifikasi dengan ukuran amplikon sebesar 600-700 bp.

ABSTRACT
Raja Ampat Islands have highly diversity of marine animals, such as Sea Cucumbers (Holothuroidea). The aim of this study is amplification based on COI gene markers of Holothuroidea which collected from the Raja Ampat Islands. DNA total was isolated from the tissue using gSYNCTM DNA Extraction Kit GS100 (Gene Aid), DNA UNP 106 specimen was amplified based on COI gene using primer forward COI : 5’-GGCATCCAGAAGTTTACATCCT-3’ and reverse COI : 5’-TTGGAGATAGGCTTCTGTGGA-3’. The results showed that UNP 106 specimen has succesfully amplified with size of amplicon is 600-700 bp.
