Analisis Polimorfisme Gen BMP-15(Bone Morphogenetic Protein-15) dengan Metode PCR-RFLP dan Hubungannya dengan Performan Reproduksi Sapi Friesian Holstein (FH)
Abstract
Penelitian ini bertujuan untuk mengetahui adanya polimorfisme gen BMP-15 (Bone morphogenetic protein-15) dengan menggunakan teknik PCR-RFLP dan hubungannya dengan performan reproduksi sapi Friesian Holstein (FH). Sampel yang digunakan dalam penelitian ini adalah 10 sapi FH betina yang diambil dari UPT. Balai Besar Pelatihan Peternakan, Songgoriti, Batu. Isolasi DNA dari darah dengan menggunakan metode salting out. Amplifikasi gen BMP-15 dilakukan dengan teknik PCR menggunakan primer forward 5’GCTCTGGAATCACAAGGGG3’ dan Primer reverse 5’AGAGATGGGGAGCGAT GAT 3’. Polimorfisme gen BMP-15 dianalisis dengan teknik PCR-RFLP menggunakan enzim restriksi HaeIII. Amplifikasi gen BMP-15 menghasilkan amplikon dengan panjang 360bp. Analisis PCR-RFLP menghasilkan dua haplotip yang menunjukkan adanya polimorfisme. Haplotip I memiliki ukuran fragmen DNA 90bp dan 190bp. Haplotip II memiliki ukuran fragmen 90bp, 190bp, 290bp, dan 350bp. Data performan reproduksi 10 sapi fertil yang digunakan adalah 3 sapi bunting dan 7 sapi memiliki data calving interval pada range 13-16 bulan, days open pada range 3-7 bulan dan 2 sapi beranak kembar. Berdasarkan data yang ada, diketahui bahwa sapi yang termasuk dalam haplotip II memiliki data performan reproduksi yang bervariasi. Berdasarkan hasil penelitian dapat disimpulkan bahwa polimorfisme gen BMP-15 yang ada, tidak menunjukkan adanya hubungan dengan performan reproduksi sapi FH.
